| Description | FUNCTION:PromotesmicrotubuleassemblyandstABIlity,andmightbeinvolvedintheestablishmentandmaintenanceofneuronalpolarity.TheC-terminusbindsaxonalmicrotubuleswhiletheN-terminusbindsneuralplasmamembranecomponents,suggestingthattaufunctionsasalinkerproteinbetweenboth.Axonalpolarityispredeterminedbytaulocalization(intheneuronalcell)inthedomainofthecellbodydefinedbythecentrosome.TheshortisoformsallowplasticityoftheCytoskeletonwhereasthelongerisoformsmaypreferentiallyplayaroleinitsstabilization.SUBCELLULARLOCATION:Cytoplasm;cytosol.Cellmembrane.Mostlyfoundintheaxonsofneurons,inthecytosolandinassociationwithplasmamembranecomponents.ALTERNATIVEPRODUCTS:8namedisoformsproducedbyalternativesplicing.Additionalisoformsseemtoexist.Isoformsdifferfromeachotherbythepresenceorabsenceofupto5ofthe15exons.Oneoftheseoptionalexonscontainstheadditionaltau/MAPrepeat.TISSUESPECIFICITY:Expressedinneurons.IsoformPNS-tauisexpressedintheperipheralnervoussystemwhiletheothersareexpressedinthecentralnervoussystem.DEVELOPMENTALSTAGE:Four-repeat(typeII)tauisexpressedinanadult-specificmannerandisnotfoundinfetalbrain,whereasthree-repeat(typeI)tauisfoundinbothadultandfetalbrain.DOMAIN:Thetau/MAPrepeatbindstotubulin.InAlzheimerdisease,theneuronalcytoskeletoninthebrainisprogressivelydisruptedandreplacedbytanglesofpairedhelicalfilamentsandstraightfilaments,mainlycomposedofhyperphosphorylatedformsofMicrotubule-associatedproteinTau.DefectsinMicrotubule-associatedproteinTauareacauseoffrontotemporaldementiaandparkinsonismlinkedtochromosome17,aswellasanumberofotherneurodegenerativediseases. |
