| Description | Caspasesplayseveralkeyrolesincellulargrowthanddevelopment,extracellularmatrixremodeling,woundhealing,homeostasis,andareimplicatedinawiderangeofdiseasesfromautoimmunedysfunctiontocancermetastasesaswell. CaspasesareCysteineASpartateProteaseswhichformafamilyofmetalloproteinasesofwhichtherearemorethanadozenmembers.Caspasesareallnormallyexpressedincellsasinactiveprecursorzymogensthatthengetactivatedviaproteolyticcleavagetoformactiveenzymecomplexes. Activationcanoccurbyseveralmeansandprogrammedcelldeath,orapoptosis,isamongthemoststudied,butthereareseveralotherssincecaspasesareinvolvedinsomanycriticalanddiseasestates. Amongthe14orsomembers,Caspase-6isparticularlyinterestingbecauseithasbeenimplicatedinplayingaroleinsomeneurodegenerativediseasesincludingAlzheimersandHuntingtonsdisease. Caspase-6hasbeenshowntocutamyloidprecursorprotein(APP),atposition720leADIngtothetoxicfragmentJcasp,whichisoneofthefragmentsfoundinamyloidplaqueswhicharebelievedtobeanindicatorofthedisease,andinHuntingtonsdisease,thespecificaminoacidsequence(IVLD586G)isrecognizedbyCaspase-6 ontheHuntingtonprotein(htt)inmicewithHuntingtonsdisease.Theproteolyticcleavageofhttliberatestoxicfragmentscontainingtheexpandedpolyglutaminetractthatareneurotoxicandthatstimulatesadditionalproteolyticactivityleadingtoapoptosisandneurodegeneration.MutationoftheCaspase-6siteinmicemodelwithAlzheimersandHuntingtonsdiseaseprovidesprotectionfromtheneuraldysfunction,suggestingacausalrelationshipbetweenCaspase-6cleavageandneurodegeneration.BiosensisispleasedtoofferarabbitpolyclonalantibodytohumanCaspase6thatreactsinparaffinembeddedimmunohistochemistryandwesternblottingforyourcontinuedresearchintoCaspase6anditspotentialrolesinbothnormalandpathologicalconditions. |
