| Description | LRRK2isamemberoftheleucine-richrepeatkinasefamily.Itroleisyetunknownbutitmayplayaroleinthephoshorylationofproteinscentraltoparkinsondiseases.LRRK2containsanankryinrepeatregion,aleucine-richrepeat(LRR)domain,akinasedomain,aDFG-likemotif,aRASdomain,aGTPasedomain,aMLK-likedomainandaWD40domain.LRRK2ispresentinthecytoplasmbutalsoassociateswiththemitochondrialoutermembrane.DefectsinLRRK2arethecauseofParkinsondisease8(PARK8).ÃÂ Parkinsondiseaseischaracterisedbybradykinesia,restingtremor,muscularrigidityandposturalinstABIlity,aswellasbyaclinicallysignificantresponsetotreatmentwithlevodopa.ThepathologyinvolvesthelossofdopaminergicneuronsinthesubstantianigraandthepresenceofLewybodies(intraneuronalaccumulationsofaggregatedproteins),insurvivingneuronsinvariousareasofthebrain.PARK8isanautosomal-dominantlate-onsetparkinsonism,characterizedbyonsetfrom50to65years,withslowprogressionandrelativelybenigncourse. |
