| Description | Myelinisamembranecharacteristicofthenervoustissueandfunctionsasaninsulatortoincreasethevelocityofthestimulibeingtransmittedbetweenanervecellbodyanditstarget.Myelinisolatedfromhumanandbovinenervoustissueiscomposedofapproximately80%lipidand20%protein,and30%oftheproteinfractionconstitutesmyelinbasicprotein(MBP).MBPisan"intrinsicallyunstructured"proteinwithahighproportion(approximately75%)ofrandomcoil,butpostulatedtohavecoreelementsofbeta-sheetandalpha-helix.MBPisamajorproteininCNSmyelinandisexpressedspecificallyinthenervoussystem.AdetailedimmunochemicalexaminationofmonoclonalandpolyclonalantibodyresponsestoMBPanditspeptideshasrevealedtheexistenceofasmanyas27antigenicdeterminants,manyofthemconformational.TopologicalmappingofthepotentialantigenicdeterminantsontoamodelofMBPsecondarystructureplacesthesedeterminantswithin11separateregionsofthemolecule,includingthoseportionsthathavebeenfoundtobeencephalitogenic.Themessageformyelinbasicproteinisselectivelytranslocatedtotheendsofthecellprocesses.Immunizationwithmyelin-associatedantigensincludingMBPsignificantlypromotesrecoveryafterspinalcordcontusioninjuryintheratmodel.FUNCTION:Is,withPLP,themostabundantproteincomponentofthemyelinmembraneintheCNS.HasaroleinboththeformationandstABIlizationofthiscompactmultilayerarrangementofbilayers.Eachsplicevariantandchargeisomermayhaveaspecializedfunctionintheassemblyofanoptimized,biochemicallyfunctionalmyelinmembrane(Bysimilarity).SUBUNIT:Homodimer(Bysimilarity).SUBCELLULARLOCATION:Myelinmembrane;peripheralmembraneprotein;cytoplasmicside.Cytoplasmicsideofmyelin.TISSUESPECIFICITY:Foundinboththecentralandtheperipheralnervoussystem. |
