DescriptionAldolasesareglycolyticenzymethatcatalyzesthereversIBLealdolcleavageoffructose1,6-bisphosphateandfructose-1-phosphatetodihydroxyacetonephosphateandeitherglyceraldehyde3-phosphateorglyceraldehyde,respectively.ThreealdolaseisozymesarefoundinmammalsspecificallyaldolasesA,B,andC,eachofwhichisencodedbyaseparategene.AldolaseAisgenerallyconsideredtobeamuscleenzyme.Northernanalysisofculturedcellssuggeststhatitispresentinbothneuronsandglia(1).AldolaseBisconsideredtobealiver-specificenzymeanditistranscriptionallyactivatedbysignalsfromhormonesanddietaryfactors(2).Intheadult,aldolaseCisthebrain-specificisozyme,withlowbutdetectableactivityinfetaltissues(1,3-6).AldolaseCshares81%aminoacididentitywithaldolaseAand70%identitywithaldolaseB.Earlierstudiesusingisozyme-specificantibodiesreportitslocationingraymatterastrocytesandcellsofthepiamater(5,8).Insituhybridizationofmousecentralnervoussystemusingisozyme-specificprobesrevealedthataldolaseAandCareexpressedincomplementarycelltypes:aldolaseAmRNAisfoundinneurons;aldolaseCmessageisdetectedinastrocytes,somecellsofthepiamater,andPurkinjecells(9).AldolaseCcaninsomesituationsbeusedasanastrocyteMarker.HoweverPurkinjecellsofthecerebellumcontainhighlevelsoftheenzyme,sotheenzymeisnottotallyastrocytespecific.
