| Description | Doublecortin(DCX,alsoknownasDoublin,Lissencephalin-X,DBCNandLis-X)wasoriginallydiscoveredsincedefectsinthegeneencodingitarecausativeofX-linkedlissencephaly,araregroupofbrainmalformationsresultinginasmoothcerebralcortexcausedbyaberrantneuronalmigrationduringdevelopment(1,2).ThenameDoublecortincomesfromtheunusuallayeringofthecortexinthisformoflissencephaly,whichappearstohaveaseconddeepcorticallayerofneurons.Thislayerconsistsofneuronswhichdidnotmigratefromthesubventricularzonetothenormalcorticallayer.Patientswiththisdefectsufferfromseizuresandmentalretardation.FourproteinsencodedbytheDCXproducebandsofabout35kDaand45kDaonWesternblots.The45kDaformisknownasLis-XAwhilethesmallerformsaregeneratedbyalternatetranscription,areallmissingthefirst81aminoacidsofLis-XA,andarereferredtoasLis-XB,Lis-XC,Lis-XD.Thereareminoraminoacidsequencedifferencesbetweenthesethreesmallerisoforms.Alloftheseproteinscontaintwoso-calledDoublecortindomains,eachabout90aminoacidslong,whicharebelievedtofunctioninbindingtomicrotubules,aC-terminalserineandprolinerichregionwhichmaybecomephosphorylatedinvivo.DCXisexpressedveryearlyinneuronaldevelopment,asneuroblastsbecomepost-mitotic,butislostasneuronsmature.DevelopingneuronsstarttoloseDCXexpressionaboutthetimethattheybegintoexpressNeuN.AntibodiestoDCXcanbeusedtoseeifneurogenesisistakingplace. |
