| Description | High-mobilitygroupproteinswerenamedoriginallysincetheyareabundandrelativelylowmolecularweightproteinswhichrunquicklyonSDS-PAGEgels.High-mobilitygroupproteinbox1(HMGB1,Amphoterin)isoneofthese.The"bx"inthenamereferstotheso-calledhighmobilitygroup(HMG)box,acompactdomaininvolvedinDNAbindingandprotein-proteininteractions.theHMGB1moleculehastwooftheseHMGdomains.Theproteinisalslocalledamphoterin,thisnamebeingderivedfromthepresenceoftwohighlychargedregionsinthemolecule,arelativelyneutrallychargedN-terminusandaverynegativelychargedC-terminus.Infactthemoleculeisveryunusuallychargedthroughout,thehumansequenceconsistingof16.7%Glutamicacid,9.3%Asparticacid,20%lysineand9.3%Arginine.HMGB1canbindTolllikereceptor4(TLR4)andtheReceptorforAdvancedGlycationEndproducts(RAGE).TLRsarecomponentsoftheinnateimmunesystem,firstrecognizedasafamilyofreceptorswhichrecognize"PathogenAssociatedMolecularPatternmolecules(PAMPs).PAMPsarecommoncomponentsofbacteriaandwhenTLRsbindtheseastronginflammatoryresponseisactivated.MorerecentlyithasbeenrecognizedthatTLRscanalsobeactivatedbyDamageAssociatedMolecularPatternmolecules(DAMPs),whichareendogenoussubstancesreleasedfromdamagedanddiseasedcellswhichalsobindtoTLRfamilyreceptorsandalsoactivateinflammation.HMGB1issuchaDAMP,bindingtoTLR4,andmuchevidencesuggeststhatHMGB1isastrongactivatorofinflammation.Interestingly,HMGB1isreleasedbynecroticcellsbutnotbyapoptoticcells(1). |
