Description	TheBiosensisGDNFRapid^TMenzyme-linkedimmunosorbentassay(ELISA)KitisasandwichELISAthatallowsthequantificationofGDNFinlessthan4hoursincellculturesupernatants,celllysates,andguineapigserumonlyifusedasdirected.Pleaserefertothekitprotocolforspecificuseinstructionsforeachsubstrateapplication. ThisELISAkitconsistsofapre-coatedanti-GDNFcaptureantibody,abiotinylatedanti-GDNFdetectionantibodyandhorserADIshperoxidase(HRP)-conjugatedstreptavidin.Theadditionofasubstrate(3,3,5,5-tetramethylbenzidine,TMB)yieldsacoloredreactionproductwhichisdirectlyproportionaltotheconcentrationofGDNFpresentinsamplesandproteinstandards. ThisGDNFELISAkitemploysarecombinanthumanGDNFstandardobtainedfromtheNationalInstituteforBIOLOGicalStandardsandControl(NIBSC)andisthereforedesignedtoaccuratelymeasurethehumanformofGDNF.Notethattheantibodiesusedinthiskitcross-reactwithguineapig,ratandmouseGDNF. GuineapigGDNFhasbeenquantifiedinserumusingtheHumanGDNFRapid^TMELISAkit,sinceguineapigGDNFproteinsharesclosestaminoacidsequencehomologywithhumanGDNF.ProteinlevelscanbeexpressedashumanGDNFequivalents.
