ThehumanHepatocyteGrowthFactor(HGF)KitisasandwichELISA.ThecaptureantibodyisamonoclonalhumanHGFantibodypre-coatedontothe96-wellstripplatesprovidedinthekit.HumantestsamplesandstandardsofknownHGFconcentrationareaddedtothesewellsandallowedtocomplexwiththeboundHGFantibody.Abiotinylatedanti-humanHGFpolyclonalantibodyisthenadded.Thisdetectionantibodybindstotheantigenthuscompletingthesandwich.Afterwashing,anenzymeAvidin-Biotin-Peroxidasecomplex(ABC)isaddedwhichbindstothesecondantibody.TheperoxidasesubstrateTMBisaddedtoinduceacolouredreactionproduct.TheintensityofthiscolouredproductisdirectlyproportionaltotheconcentrationofHGFpresentinthesamples.Thepurposeofthiskitisthein-vitroquantitativedeterminationofhumanHGFinsamplessuchassera,plasma,andcellculturesupernates.Thiskithasbeenconfiguredforresearchuseonlyandisnottobeusedindiagnosticorclinicalprocedures.
