Description	TheBiosensisMouseandRatproNGFRapid^TMenzyme-linkedimmunosorbentassay(ELISA)KitisasandwichELISAthatallowsthequantificationofrodentfull-lengthproNGFproteininlessthan4hoursincellculturesupernatantsandcelllysatesonlyifusedasdirected.Pleaserefertothekitprotocolforspecificuseinstructionsforeachsubstrateapplication. ThisELISAkitcontainsarecombinantmouseproNGFstandardexpressedinE.coliandconsistsofapre-coatedanti-proNGFcaptureantibody,abiotinylatedanti-proNGFdetectionantibodyandhorserADIshperoxidase(HRP)-conjugatedstreptavidin.Theadditionofasubstrate(3,3,5,5-tetramethylbenzidine,TMB)yieldsacoloredreactionproductwhichisdirectlyproportionaltotheconcentrationofproNGFpresentinsamplesandproteinstandards. ThisELISAkitisexpectedtodetectratproNGFduetohighdegreeofhomology(96%)withmouseproNGFbasedonaminoacidsequence,andtheABIlityofthiskitindetectingproNGFinratPC12celllysates.IntheabsenceofatrueratproNGFstandard,resultsmaybeexpressedas"mouseproNGFequivalents". ThisELISAkitshowsonly20%reactivitywiththehumanformofproNGFandisthereforenotsuitabletoquantifyhumanproNGF.Nocross-reactivitywasobservedwithmaturemouseNGFandfull-lengthproBDNFwhentestedinassaybuffer.TheantibodiesusedinthisELISAkitbindepitopeswithinthepro-domain(capture)andmaturedomain(detection)oftheprotein,thusthisELISAassaydoesnotdetectthepro-domainpeptide. Thiskithasnotbeentestedforotherapplications.SufficientamountofproNGFstandardissuppliedtoallowforspike-andrecoveryexperimentsinordertovalidatethisELISAassayforothersamplematricesifrequired.Thiskithasbeenconfiguredforresearchuseonlyandisnottobeusedindiagnosticorclinicalprocedures.
