Description	TheBiosensisNT3Rapid^TMenzyme-linkedimmune-sorbentassay(ELISA)KitisasandwichELISAthatallowsthespecific,fastandreliablequantificationofNT3inlessthan4hoursincellculturesupernatantsandhumanplasma(EDTAandcitrate)onlyifusedasdirected.Pleaserefertothekitprotocolforspecificuseinstructionsforeachsubstrateapplication,inparticularhumanplasma.AccuratequantificationofNT3inhumanplasmarequiresaheterophilicantibody(HA)blockerwhichcanbepurchasedseparately(BL-004-500). ThisELISAkitconsistsofapre-coatedpolyclonalanti-NT3captureantibody,abiotinylatedmonoclonalanti-NT3detectionantibodyandhorserADIshperoxidase(HRP)-conjugatedstreptavidin.Theadditionofasubstrate(3,3,5,5-tetramethylbenzidine,TMB)yieldsacoloredreactionproduct,whichisdirectlyproportionaltotheconcentrationofNT3presentinsamplesandproteinstandards. ThisNT3Rapid^TMELISAkitemploysarecombinanthumanNT3standard.NT3ishighlyconservedandnearlyidenticalinmanyspecies.Theassay"scaptureanddetectionantibodiesdetectNT3fromotherspeciesincludingmouse,ratandmonkey,thusitisexpectedthatthisNT3Rapid^TMELISAwillalsoreactwiththosespeciesandpossIBLymorebecauseoftheconversationoftheimmunogenused. TheantibodiesusedinthiskitbindtoepitopeswithinthematuredomainofNT3.Thus,thisELISAkitmaydetectthefull-lengthpro-formofNT3. Thiskithasnotbeentestedforotherapplications.SufficientamountofNT3standardissuppliedtoallowforspike-andrecoveryexperimentsinordertovalidatethisELISAassayforothersamplematricesifrequired.Thiskithasbeenconfiguredforresearchuseonlyandisnottobeusedindiagnosticorclinicalprocedures.
