Description	TheBiosensisNGFRapid^TMenzyme-linkedimmunosorbentassay(ELISA)KitisasandwichELISAthatallowsthequantificationofhumanNGFinlessthan3hoursincellculturesupernatants,serum,plasma(citrate)andbrainextractsonlyifusedasdirected.Pleaserefertothekitprotocolforspecificuseinstructionsforeachsubstrateapplication,inparticularserumandplasmasamples. ThisELISAkitconsistsofapre-coatedmousemonoclonalanti-NGFcaptureantibody,abiotinylatedanti-NGFdetectionantibodyandhorserADIshperoxidase(HRP)-conjugatedstreptavidin.Theadditionofasubstrate(3,3,5,5-tetramethylbenzidine,TMB)yieldsacoloredreactionproductwhichisdirectlyproportionaltotheconcentrationofNGFpresentinsamplesandproteinstandards.AhumanNGFpositivecontrol(QCsample)isprovidedtoassureconsistentassayperformance. ThisNGFELISAkitisdesignedtomeasurehumanNGFandthusemploysarecombinanthumanNGFstandardapprovedbytheWorldHealthOrganization(WHO).DuetoahighdegreeofNGFsequencehomology,theantibodiesusedinthiskitwillalsodetectNGFfromnumerousotherspeciesincludingmouseandrat! TheantibodiesusedinthisELISAkitbindepitopeswithinthematuredomainoftheproteinandthereforecanrecognizeboththepro-andthematureformofNGF.However,internalvalidationdatademonstrates<0.1% cross-reactivity (determined at 25 ng/mL, diluted in assay buffer) with human proNGF protein, suggesting the preferential quantification of mature NGF over full-length human proNGF NotethataccurateNGFquantificationinhumancitrate-plasmarequirestheadditionofHeterophilicAntibodyBlockerBL-005-500providedinthekit,andavailableforpurchaseseparately.
