DescriptionTheBiosensisNGFR/p75ECDRapidTMenzyme-linkedimmunosorbentassay(ELISA)KitisasandwichELISAthatallowsthequantificationoftheextracellulardomain(ECD)ofhumanp75neurotrophinreceptorinlessthan4hoursinurine,serum,citrate-plasma,CSFandcellculturesupernatantsonlyifusedasdirected.Pleaserefertothekitprotocolforspecificuseinstructionsforeachsubstrateapplication.

Notethatcaptureanddetectionantibodieshavebeenoptimizedforincreasedassaysensitivity,andreagentsfrompreviouslotsshouldnotbeused.Thisoptimizedhumanp75ECDELISAassaydemonstratesanincreasedsensitivity,loweringtheLODabout8-foldtothepreviousassayversion.ItrepresentsthemostsensitiveNGFR/p75ECDELISAassayavailabletoday,withadetectionlimitofatleast6pg/mL,andaquantificationrangeof7.8-500pg/mL.

ThisELISAkitconsistsofapre-coatedmousemonoclonalanti-p75ECDcaptureantibody,abiotinylatedmousemonoclonalanti-p75ECDdetectionantibodyandhorserADIshperoxidase(HRP)-conjugatedstreptavidin.Theadditionofasubstrate(3,3’,5,5’-tetramethylbenzidine,TMB)yieldsacoloredreactionproductwhichisdirectlyproportionaltotheconcentrationofp75ECDpresentinsamplesandproteinstandards.Ahumanp75ECDpositivecontrol(QCsample)isprovidedtoassureconsistentassayperformance.

ThisNGFR/p75ECDELISAkitemploysanin-houserecombinanthumanp75ECD-Fcchimeraasstandardthathasbeenvalidatedagainstanexternallyavailablep75ECD-Fcprotein.Whilethereisacurrentlackofacommerciallyavailable,truehumanp75ECDstandard,thisELISAkitallowsquantificationofhumanp75ECDasp75ECD-Fchumanequivalents.PleasenotethattheantibodiesusedinthisELISAdonotcross-reactwithmousep75ECD.
